Line or instrument level?

I am using Helix preamps running into the effects return of a Rivera Chubster 55, by passing the Rivera's preamp.  Should my global settings be on line or instrument?

I would think an amp's effects loop would be looking for line level signals, like from rack gear.

Exactly...instrument level is what goes into the normal guitar input of an amp.
