what is the diference of S/PDIF

Every time you go from analog to digital or digital to analog there's a conversion (ADC/DAC) from a voltage based signal to a bunch of 0's and 1's or vice versa. Each conversion takes time (latency) and slightly degrades the signal, though whether or not that degradation is perceptible is a topic of some debate.

Guitar to Helix = 1 DAC

Helix to Line Out (XLR or 1/4") = 1 DAC

S/PDIF eliminates that second DAC.

FWIW - I can't tell a difference in audio quality, and consider the added latency to be negligible. YMMV.

You may want to read this if you are not familiar with S/PDIF:

hope this helps / makes sense.
