If I am plugging into an amp and playing live should I therefore turn off the amp & cab simulator?

I can't get the Pitch Wham to sound usable at all. I assigned it to the Expression 2, and changed the heel & toe + - to many different combinations. Basically I just get a high pitched shrill when I depress the pedal. How to dial it in?
