So - on further investigation, looks like a bug related to Low Latency Monitoring. Low Latency Monitoring off? Everything works as expected and I hear patch changes as I make them. Low Latency Monitoring on? I can twiddle Helix Native's knobs until the cows come home, I don't hear any changes at all. I'm assuming this is a bug (unless the view is this is a well known "feature" and everything is working "by design"...)

Hi, N00b Native user here. With Studio One 6 Professional, Helix Native is ignoring all patch changes and patch edits unless I reset the Track's input (e.g. from mono to stereo). This makes Helix Native largely useless. I don't have any other Studio One issues so I'm wondering if this is a bug - are any other Studio One users having similar problems? (I have raised tickets with L6 and PreSonus.)
