The tuner inputs you can use are Guitar, Variax (and Aux for Helix; see tuner settings)...

for you... My Variax, guitars and basses can be tuned correctly with helix. (And other tuners ;-) ) Check the Helix tuner settings: In(put) for example. Check the reference (440Hz)...

Welcome to the forum! You cannot assign an octaver (or any other effect) to single strings (or notes) on a 'regular' guitar. You can use alternative tunings with a Variax.

Maybe you hear something like a a very thin delayed sound of a repressed metal spring (on a footswitch). But I don't think so. Don't think of it anymore, or you'll get a tinnitus ;) Yes. They do. If it bothers you too much, draw a support ticket.

These discussions reappear very often... Many players seem incessantly looking for an amplifier sound, not for a mic'd amplifier sound. And understanding this means to listen to it at as the audience (or the soundguy) does...
